Non-immunogenic, Non-human Proteins

Amunix stands for ‘non-immunogenic (protein) therapeutics’. Typical proteins are globular and require a hydrophobic core for folding. 

One way to make a protein less immunogenic is to maximize it’s entropy, by maximizing flexibility and minimizing structure. This approach has led to our XTEN technology for serum half-life extension. XTEN is a long, linear, unstructured polymeric protein tail that is genetically fused to an existing pharmaceutical protein, such as G-CSF, IFN or hGH. Because the apparent molecular weight of such an unstructured protein chain is about 15-fold larger than its actual molecular weight, the serum half-life of the protein is greatly increased. Because XTEN is added genetically, it does not require the chemical conjugation and re-purification steps that are required for PEG.  Compared to PEGylation, the manufacturing process is greatly simplified and the product is homogeneous.
